        Phosphotungstic Acid Hematoxylin Stain

        Principle:

        Mature hematin reacts with tungsten to form blue sediment which strongly combines with tissue. Reddish brown portion is stained by Phosphotungstic Acid.

        Methods:

        1. Put tissue into xylene. In subsequent step, put the tissue in tap water.

        2. Add distilled water into 5% iron alum and mordant for 30 minutes.

        3. Rinse with distilled water after washing in tap water.

        4. Oxidize the smear in 2.5% potassium permanganate and rinse with tap water.

        5. Place in 2.5% oxalate till the smear turns white.

        6. Rinse in tap water, distilled water. Then, dry with absorbent paper.

        7. Place in PTAH stain for 12~24hours, or incubate for an hour.

        8. Differentiate in 95% alcohol for 3~5 seconds.(As soon as possible)

        Rapidly dehydrate; clear and mount.

         

        Expected Results:

        Rhabdium, Glia, Fibrin, Nuclei……..……..Blue

        Collagen fibers……………………………..Reddish Brown

         

        Precaution:

        1. Do not rinse with water after staining with Phosphotungstic Acid Haematoxylin. Quickly rinse in 95% alcohol, because alcohol or water will wash red color away.

        2. Do not overstain the smear, since Phosphotungstic Acid Haematoxylin is progressive stainning reagent. Examine under a microscope in certain period.

        3. The procedure of staining is to be operated in a staining jar.
